ABSTRACT

A 23-year-old patient presented with complaints of redness, pain, photophobia, and blurred vision in the right eye 15 days after she received the third dose of BNT162b2 vaccination. Ocular examination revealed 2+ cellular reactions in the anterior chamber and mutton fat keratic precipitate with no vitritis or retinal alterations. Active uveitis findings regressed with corticosteroid and cycloplegic eye drops. We present a case of unilateral granulomatous anterior uveitis following the BNT162b2 vaccination, with no etiologic factor in uveitis work-up and no previous history of uveitis before vaccination. This report demonstrates a potential causal association of coronavirus disease 2019 (COVID-19) vaccine with granulomatous anterior uveitis.

ABSTRACT

In order to study the analgesic effect of Shaoyao Gancao Decoction, this paper discussed material basis and mechanism from the perspective of macromolecules in traditional Chinese medicine. Inspired by the phenomenon of turbidity after boiling Chinese medicine, this experiment took Shaoyao Gancao Decoction as the research object to study the formation process of precipitation during boiling. The results showed that aggregates with a certain shape were formed in the solvent during the boiling process, and the precipitate was obtained by standing and centrifuging. Analysis found that the precipitation was mainly composed of small molecules such as paeoniflorin, albiflorin, liquiritin, glycyrrhizic acid, isoliquiritin and gallic acid, and macromolecules such as protein and polysaccharide. The composition of precipitate was consistent with that of Shaoyao Gancao Decoction, but the analgesic effect of Shaoyao Gancao Decoction after removing the precipitate was significantly reduced. Based on these results, we isolated small molecular compounds, polysaccharides and protein from Shaoyao Gancao Decoction and their contents are 60.4, 700.7 and 207.2 mg·g-1 respectively. We get the ratio, polysaccharide: small molecule = 11.6∶1, protein: small molecule = 3.4∶1, the precipitate is prepared in the state of boiling. The characterization results showed that the particle size of the precipitate will change significantly after co-heating, and the content determination results showed that the content of the six small molecular compounds which was free in solvent was significantly reduced after the formation of the precipitate. The acetic acid writhing experiment proved that the precipitate has a good analgesic effect, and effectively reduced the levels of inflammatory factors prostaglandin E2 and nitric oxide, and increased the level of anti-inflammatory factor interleukin-10. These results proved that the precipitate in Shaoyao Gancao Decoction is an important material basis for analgesic effect, and macromolecules such as protein and polysaccharide are the main components of the precipitate. The study of macromolecules in the precipitate of Shaoyao Gancao Decoction not only provides new ideas and methods for elucidating the pharmacodynamic material basis of Shaoyao Gancao Decoction, but also provides a reference for analyzing the scientificity of traditional decoction.

ABSTRACT

OBJECTIVE To study the content changes of 5 chemical compositions in water extract and ethanol precipitate of different processed products of Psoralea corylifolia, and to preliminarily evaluate its hepatotoxicity. METHODS The water extracts from crude product of P. corylifolia and processed products by Leigong method, running water rinsing method, and salt stir-frying method were prepared, as well as the ethanol precipitates of processed products by Leigong method and salt stir-frying method were prepared. The contents of psoralenoside, isopsoralenoside, psoralen, isopsoralen and bakuchiol were determined by high- performance liquid chromatography and compared. The median lethal concentration (LC50) and maximum non-lethal concentration (MNLC) of each sample to wild-type zebrafish juveniles were calculated after 72 h of treatment with different concentrations of water extracts from raw product and processed products by running water rinsing method, Leigong method and salt stir-frying method, different concentrations of ethanol precipitates from processed products by Leigong method and salt stir-frying method, and the acetaminophen was used as the positive control. The basic morphology of wild-type zebrafish juveniles and the liver phenotype of transgenic zebrafish juveniles were observed after 72 h of treatment with the above samples (MNLC). Pearson correlation analysis was used to evaluate the correlation between component content and hepatotoxicity. RESULTS Compared with the water extract of raw products, the contents of psoralenoside and isopsoralenoside in the water extract of different processed products were generally decreased (P<0.05), while the contents of psoralen, isopsoralen and bakuchiol in the ethanol precipitate of Leigong method and salt stir-frying products were significantly increased (P<0.05). The LC50 of water extracts of crude product and processed products by running water rinsing method, Leigong method, salt stir-frying method, and ethanol precipitates of processed products by Leigong method and salt stir- frying method were 2.45, 5.00, 5.38, 1.55, 2.36, 0.64 g/L (calculated by crude drug), and MNLC were 2.21, 4.53, 5.02, 1.37, 2.13, 0.53 g/L (calculated by crude drug). Compared with the blank control group, the zebrafish juveniles in each sample treatment group showed different degrees of deformity, the liver relative fluorescence intensity was significantly weakened (P<0.05 or P< 0.01). Fat-soluble components such as bakuchiol, isopsoralen and psoralen were highly correlated with liver fluorescence intensity (R 2>0.7). CONCLUSIONS The processed products of P. corylifolia mainly compose of psoralenoside and isopsoralenoside after water extraction, the contents of psoralen, isopsoralen and bakuchiol increase after alcohol precipitation, and the hepatotoxicity is positively correlated with the contents of liposoluble compositions in P. corylifolia.

OBJECTIVE@#To investigate the perinatal outcome and risk factors of precipitate labor in term primipara.@*METHODS@#A total of 6951 full-term singleton primiparas with cephalic vaginal delivery in Women's Hospital, Zhejiang University School of Medicine from January 2020 to December 2020 were enrolled, among whom 381 cases of precipitate labor were classified as the precipitate labor group and 762 cases of normal labor were randomly selected as the control group. The perinatal outcomes of the two groups were compared, and the risk factors of precipitate labor were analyzed by multivariate logistic regression.@*RESULTS@#The incidence of precipitate labor in full-term, singleton pregnancy and cephalic primiparas was 5.48% (381/6951). The durations of the first and second stages of labor in the precipitate labor group were significantly shorter than that in the control group ( P<0.01); while there was no significant difference in the duration of the third stage of labor between the two groups ( P>0.05). Compared with the control group, the incidence of soft birth canal laceration in the precipitate labor group was increased ( P<0.01). However, there was no significant difference in postpartum hemorrhage and neonatal related perinatal outcomes between the two groups (all P>0.05). Multivariate logistic regression analysis showed that maternal height ( OR=1.038, 95% CI: 1.010-1.067, P<0.01), gestational age at delivery ( OR=0.716, 95% CI: 0.618-0.829, P<0.01), late miscarriage ( OR=1.986, 95% CI: 1.065-3.702, P<0.05), membrane rupture before labor ( OR=1.802, 95% CI: 1.350-2.406, P<0.01), labor induction by transcervical balloon ( OR=3.230, 95% CI: 2.027-5.147, P<0.01), labor induction by propess ( OR=2.332, 95% CI: 1.632-3.334, P<0.01) and labor induction by oxytocin ( OR=0.291, 95% CI: 0.219-0.386, P<0.01) were independently associated with precipitate labor.@*CONCLUSIONS@#The incidence of precipitate labor in full-term, singleton pregnancy was not low. Precipitate labor could lead to a significant increase in perineal laceration. Maternal height, history of late miscarriage, membrane rupture before labor and labor induction by transcervical balloon, labor induction by propess are risk factors, while labor induction by oxytocin and late gestational time of delivery are protective factors for precipitate labor in term primipara.

ABSTRACT

The high performance liquid chromatography(HPLC) characteristic chromatogram of Xiaoer Ganmaoning Oral Liquid(oral liquid for short) was established. The medicinal materials corresponding to characteristic peaks, their index components and ranges of similarity with the reference chromatograms were clarified. The similarity between the characteristic chromatograms of 10 batches of the oral liquid and the reference chromatogram was higher than 0.994. Eighteen characteristic peaks were identified, which were derived from different medicinal materials including Scutellariae Radix, Arctii Fructus, Lonicerae Japonicae Flos, Gardeniae Fructus and Forsythiae Fructus. Further, 11 characteristic peaks were assigned by the comparison with reference substances as chlorogenic acid, neochlorogenic acid, cryptochlorogenic acid, isochlorogenic acid A, isochlorogenic acid C, baicalin, baicalein, wogonin, scutellarin, forsythiaside A and arctiin. Also, the characteristic chromatogram of precipitate in the oral liquid was established, and the similarity between characteristic chromatograms of 10 batches of the precipitate and the reference chromatogram was higher than 0.940. The 14 characteristic peaks originating from the precipitate and those from the oral liquid were consistent in retention time, and the content of all index components in the precipitate was lower than 5% of that in the oral liquid. Moreover, the stability of precipitate during the accelerated stability test was explored with filtration and Matlab-based image sensory evaluation. The precipitate mass and precipitation degree both increased over the stability test duration significantly. The stability of the oral liquid was used as a model system in this study to establish the integrated quality control system which related to medicinal materials, preparations and precipitate with HPLC characteristic chromatograms and image sensory evaluation, which lays a foundation for the exploration of the quantity value transfer of the oral liquid.

ABSTRACT

l objetivo de este estudio fue analizar el precipitado formado por la interacción de dos disoluciones acuosas: una de hipoclorito de sodio al 5.25% y otra de gluconato de clorhexidina al 2%, por medio de cromatografía de capa fina (TLC) y un análisis detallado de espectroscopia de resonancia magnética nuclear (RMN-¹H a 600 MHz y RMN-¹³C a 100 MHz) en una y dos dimensiones. Para esto, se establecieron 3 grupos de estudio: el Grupo A correspondiente a gluconato puro de clorhexidina que fue liofilizado y secado al vacío, el Grupo B: los precipitados obtenidos al combinar 2 ml de la disolución de gluconato de clorhexidina con 2 ml de la disolución de hipoclorito de sodio y el Grupo C, una muestra comercial de PCA (4-Cloroanilina 98%,). El sólido correspondiente al grupo B, fue lavado, centrifugado y seco en estufa de vacío sin calentamiento por más de 72 horas. Una vez seco, se corrieron placas de capa fina (TLC) en diversas mezclas de elución, y se encontró que el precipitado consistía de una mezcla compleja de sustancias similares a la clorhexidina y que no poseía PCA. Los análisis de espectroscopia de resonancia magnética nuclear mostraron que la señal del carbono base del grupo amino de la PCA, a δ/146.5 ppm (grupo C), no se encontraba en el espectro de ¹³C de las muestras del grupo B lo que implica, la ausencia de PCA en la muestra B. El análisis del grupo B por medio de la misma técnica, mostró una mezcla compleja de señales que corresponden probablemente, a estructuras similares a la clorhexidina y a potenciales derivados aromáticos con una estructura similar a esta, nuevamente, no se encuentran evidencias de PCA.


he aim of this study was to analyze the precipitate formed by the mixture of 5,25% sodium hypochlorite (NaOCl) and 2% chlorhexidine (CHX) through thin layer chromatography and nuclear magnetic resonance spectroscopy (RMN-¹H, 600 MHz and RMN-¹³C to 100 MHz) 1D and 2D spectra. Thus, the following groups were established: Group A corresponds to a pure freeze-dried chlorhexidine gluconate , Group B made-up by a combination of 2ml of chlorhexidine 2% and sodium hypochlorite 5.25% and Group C was a commercial sample of PCA (4-Chloroaniline 98%). The samples of group B were rinsed with distillated water and spinned during 15 minutes at 25°C, the supernatant was eliminated by vacuum and vacuum chamber for 72 hours without heating. Finally, the solid was grinded and dried in vacuum chamber for 24 hours without heating. Thin layer chromatography, shows that sample B were composed by more than one chemical substance and Chlorexidine, the RMN-¹³C showed that the signal of the amino group characteristic of PCA appears down field (δ/146.5 ppm) in C group, meanwhile in group B appears up field (δ/129ppm), which demonstrates the absence of PCA during the process. The analysis of Group B by RMN-¹³C results also, in different signals of low intensity that correspond to similar structures to chlorhexidine and potential aromatics derivatives with similar characteristics structures to chlorhexidine.

ABSTRACT

El objetivo del presente estudio fue comparar el método de precipitación con el método de fórmula de Friedewald para la determinación de colesterol LDL en caninos. Para ello, se tomaron muestras de sangre de 185 caninos adultos en estado de ayuno de diferente raza y sexo. Se determinaron los niveles de colesterol LDL mediante el método precipitado y posteriormente con el método de Friedewald. Los resultados fueron analizados estadísticamente mediante ANOVA de una vía. El método precipitado reportó valores en mg/dl de: 52,40 promedio; 2,66 mínimo; 132,67 máximo; 130,01 rango y 24,29 de desviación estándar. Por su parte, los valores del método de Friedewald en mg/dl fueron: 65,19 promedio; 4,55 mínimo; 184,20 máximo; 179,65 rango y 31,51 de desviación estándar. El valor de p en el test F fue menor a 0.05, indicando diferencia estadísticamente significativa entre los dos métodos analizados con un nivel de confianza del 95,0%. En conclusión se recomienda utilizar el método precipitado para la determinación de los niveles de colesterol LDL en caninos.


The objetive of this study was to compare the precipitation method with the method of Friedewald formula for the determination ofLDL cholesterol in dogs. For that, blood samples of 185 overnight fasted adult dogs of different breed and sex were obtained. Blood levels of LDL cholesterol were determined by the precipitation method and later by the Friedewald method. The results were analyzed using one way ANOVA. The precipitate method reported values in mg/dl of: 52.40 average, 2.66 minimum, 132.67 maximum, 130.01 range and 24.29 of standard deviation. For its part, the Friedewald equation values in mg/dl were: 65.19 average, 4.55 minimum, 184.20 maximum, 179.65 range and 31.51 of standard deviation. The p-value in the F test was less than 0.05, indicating statistically significant difference between the two methods analyzed with a confidence level of 95.0%. In conclusion it can be used the precipitated method for determining LDL cholesterol levels in dogs.

ABSTRACT

This article was aimed to study macroporous resin adsorption kinetics for effective extraction of water ex-tracting with alcohol precipitating in cicada slough. PT, APTT and the coagulation-fibrinolysis dynamic figure were taken as main indexes, which were combined with static and dynamic tests, to select the best macroporous resin to separate and purify the extraction. Adsorption kinetics curve was drawn to fit the adsorption kinetics model. The re-sults showed that NKA-9 macroporous resin was more effective in separating and purifying effective extraction than others. The adsorption dynamic behavior was well described by the pseudo-second-order kinetics equation. It was concluded that the adsorption rate was mainly controlled by the intraparticle diffusion.

Introduction: Irrigants play an essential role in the successful debridement and disinfection of pulp space. Various combination of irrigants used during root canal treatment enhance their efficacy, but some form precipitates which affects the diffusion of intracanal medicaments and the seal of the obturated root canal. Aim: To evaluate the combination of various irrigants whether it forms the precipitate and also to quantify the amount of precipitate formed. Materials and Methods: Five different irrigants were taken in ten test tubes each having the combination of two irrigants and observed for 2 min for precipitate formation. Results: One-way-ANOVA test revealed more amounts of precipitate with 2% Chlorhexidine (CHX) 5% Tulsi followed by 2% CHX, 5% Neem and 3% Sodium hypochlorite 2% CHX. Conclusion: Precipitate formation is seen more with the 2% CHX-5% Neem followed by 2% CHX-5% Tulsi when compared with the control group. When these combinations of irrigants were used, intermediate flushes of saline or distilled water reduced the amount of precipitate.

ABSTRACT

The purpose of this study was to compare the different canal irrigation methods to prevent the formation of precipitate between sodium hypochlorite (NaOCl) and chlorhexidine (CHX). Extracted 50 human single-rooted teeth were used. The root canals were instrumented using NiTi rotary file (Profile .04/#40) with 2.5% NaOCl and 17% EDTA as irrigants. Teeth were randomly divided into four experimental groups and one control group as follows; Control group: 2.5% NaOCl only, Group 1: 2.5% NaOCl + 2% CHX, Group 2: 2.5% NaOCl + paper points + 2% CHX, Group 3: 2.5% NaOCl + preparation with one large sized-file + 2% CHX, Group 4: 2.5% NaOCl +95% alcohol+ 2% CHX. The teeth were split in bucco-lingual aspect and the specimens were observed using Field Emission Scanning Electron Microscope. The percentages of remaining debris and patent dentinal tubules were determined. Statistical analysis was performed with one-way analysis of variance (ANOVA). Energy Dispersive x-ray Spectroscopy was used for analyzing the occluded materials in dentinal tubule for elementary analysis. There were no significant differences in percentage of remaining debris and patent tubules between all experimental groups at all levels (p > .05). In elementary analysis, the most occluded materials in dentinal tubule were dentin debris. NaOCl/CHX precipitate was detected in one tooth specimen of Group 1. In conclusion, there were no significant precipitate on root canal, but suspected material was detected on Group 1. The irrigation system used in this study could be prevent the precipitate formation.

ABSTRACT

Objective To isolate and purify human coagulation factor Ⅶ from Cohn fraction Ⅲ precipitate.Methods The purification procedure of human factor Ⅶ from Cohn fraction Ⅲ precipitate involves dissolving fraction Ⅲ,absorbing factor Ⅶ onto barium citrate and eluting,ammonium sulfate fractionation and DEAE-Sepharose Fast Flow ion exchange chromatography,and Sephadex G-100 gel filtration chromatography.Results 10.1mg purified FⅦ was obtained from 400g Cohn fraction Ⅲ precipitate.The purified FⅦ has a specific clotting activity of 1775.8U/mg and the overall yield of FⅦ specific clotting activity is 17.6% of the starting material.The purity of FⅦ was judged by SDS-PAGE and there was only one protein band on the gel.Conclusion The procedure of purifying Ⅶ from Cohn fraction Ⅲprecipitate is established with satisfactory purity.
